High-throughput Assay for Simultaneously Detecting Multiple PDZ Domain Interactions

Redwood City, Calif. February 18, 2003—Panomics, Inc. introduces the PDZ Domain Array for determining whether a protein of interest binds to human PDZ domains. Unlike traditional methods such as co-immunoprecipitation, the PDZ Domain Array facilitates assessment of binding to 28 PDZ domains in one hybridization. Moreover, this array-based assay identifies interactions between a ligand and PDZ domains that remain folded in active conformations.

The assay is simple: the protein of interest is expressed in bacteria, then the extract is incubated with the array membrane. The protein interactions literally take place on the array membrane, and can be easily visualized by HRP-based chemiluminescence detection. The product comes with everything needed: two array membranes, ligand expression vector, assay buffers, positive control, and detection reagents.
