New Array for Characterizing Activities of 96 Transcription Factors

Redwood City, Calif. June 19, 2002—Panomics, Inc. introduces the Protein/DNA Array II—the second edition of our array for high-throughput functional analysis of eukaryotic transcription factors (TFs). This new array represents a significant improvement over cumbersome gel mobility-shift assays, which only permit the characterization of one TF at a time. With the Protein/DNA Array II, the activities of 96 TFs can be characterized simultaneously. The Protein/DNA Array I, which includes 54 TF consensus sequences, is also available.

The procedure for both arrays is simple and straightforward, requiring no special equipment for detection. The first step is to prepare nuclear extracts from the two samples being compared. When the probe mix is added to the nuclear extracts, DNA/protein complexes form, representing the TFs of interest. The unbound probe is removed by washing; then, the DNA is separated from the TFs by gel electrophoresis. Finally, the formerly TF-bound DNA sequences are hybridized to Array membranes, and the signal is detected by HRP-based chemiluminescence. All reaction and detection reagents are included.
